Description

ACE-031 is a recombinant fusion protein compound consisting of the extracellular domain of the Activin Type IIB receptor (ActRIIB) and the Fc region of human IgG1. The substance functions as a decoy receptor binding myostatin and related ligands (Activin A, GDF-11) and thereby preventing their signal action at the actual ActRIIB receptor.

Per study reports ACE-031 blocks the myostatin pathway - myostatin is the central negative regulator of muscle growth. Studies in animal models with myostatin knockout (e.g., Belgian Blue cattle, Schwarzenegger mice) show dramatic muscle hypertrophy. ACE-031 mimics this knockout effect pharmacologically.

Acceleron Pharma developed ACE-031 for Duchenne muscular dystrophy (DMD) research in clinical Phase II. Studies showed significant muscle hypertrophy but were stopped due to severe side effects (nosebleeds, telangiectasias).

In the recreational community ACE-031 is a young and very potent anabolic compound, but cardiac and vascular safety data isn’t complete. The substance blocks not only myostatin but also related ligands, explaining off-target effects.

Risks & Safety

ACE-031 is a young and very potent anabolic compound whose safety data is less complete compared to the classical healing peptides. As an Fc fusion protein with a long half-life, the action persists for roughly two weeks.

Observed in research and user reports:

  • Injection site: mild redness, pressure or transient irritation at the injection site with subcutaneous use.
  • Vascular effects: nosebleeds and telangiectasias (dilated small blood vessels) were documented in the clinical Phase II studies - the reason the studies were stopped. This is the central observation point of this compound.
  • Off-target binding: ACE-031 blocks not only myostatin but also related ligands such as Activin A and GDF-11, which explains the vascular off-target effects.
  • Long half-life: due to the Fc portion, the substance remains active for roughly 10-15 days, so a reaction that has occurred subsides correspondingly slowly.
  • No long-term data: cardiac and vascular safety data is not complete, and robust long-term human studies do not exist.
