Mazdutide (also IBI362 or LY3305677) is an oxyntomodulin analog acting simultaneously on GLP-1 and glucagon receptors. Structurally, it is a synthetic analog of the naturally occurring hormone oxyntomodulin, which addresses both receptor systems in the human body. In research models, the dual activation is associated with a combination of appetite-regulating effects (via GLP-1) and increased energy expenditure (via glucagon).
The compound originates from a collaboration between Innovent Biologics and Eli Lilly, with Innovent leading development in China. Phase 2 studies observed body weight reductions of up to 15.4 percent after 24 weeks. Phase 3 data from the Chinese market is currently available; the substance was approved there for use in 2025.
Pharmacokinetically, Mazdutide is designed for weekly subcutaneous use in research protocols. The half-life is in the range of approximately seven days, supporting the weekly frequency. Maximum plasma concentration is typically reached after around 72 hours.
Within the GLP-1/glucagon class, Mazdutide is one of the few compounds directly modeled on the natural hormone oxyntomodulin. This makes the compound interesting for comparative research using natural oxyntomodulin as a reference.

Risks & Safety
Mazdutide is considered generally well tolerated per study reports, although as a GLP-1/glucagon dual agonist the documented profile is shaped by gastrointestinal effects and is dose-dependent in the Phase 2 data.
Observed in studies and user reports:
- Gastrointestinal effects: nausea, vomiting, diarrhea and constipation are the most commonly documented observations, particularly at the start and with overly rapid dose escalation. Slow titration reduces the incidence.
- Appetite and fluids: the reduced appetite can lead to decreased food and fluid intake, and the literature describes an elevated risk of dehydration.
- Heart rate: the literature documents mild increases in heart rate attributed to the glucagon component.
- Muscle mass and injection site: weight loss involves a portion of muscle mass alongside fat mass, and mild irritation at the injection site is documented.
- No long-term data: most of the available evidence comes from the Chinese market, and robust data on very long-term use do not yet exist.
