Description
CJC-1295 with DAC is the long-acting variant of the CJC-1295 molecule. The Drug Affinity Complex (DAC) is a maleimide group that covalently binds to plasma albumin, extending the half-life of the substance from hours to several days. This fundamentally distinguishes CJC-1295 with DAC from the DAC-free variant (Mod GRF 1-29), which has a much shorter duration of action.
Pharmacologically, CJC-1295 is a modified analog of natural growth hormone-releasing hormone (GHRH 1-29). It binds to GHRH receptors on the anterior pituitary and stimulates GH release. Through DAC modification and albumin binding, the substance remains active in plasma for several days. In research studies, GH and IGF-1 elevations of up to six days following single dosing have been observed.
In research models, CJC-1295 with DAC produces a continuous elevation of GH plasma levels rather than the pulsatile release characteristic of the DAC-free variant. This makes the compound interesting for setups investigating sustained GH axis activation. Simultaneously, this means loss of natural pulsatility, which is debated in research discussions as a trade-off against the DAC-free variant.
The typical application frequency in research protocols is once or twice subcutaneously per week.

Risks & Safety
CJC-1295 with DAC is considered well tolerated per study reports. As a GHRH analog, the compound stimulates the body’s own GH secretion - the DAC modification, however, produces a continuous rather than pulsatile GH elevation over several days.
Observed in research and user reports:
- Injection site: mild redness, pressure or transient irritation at the injection site is the most common observation with subcutaneous use.
- GH-typical effects: as is common with GH secretagogues, the literature describes transient water retention as well as numbness or tingling in the hands (carpal-tunnel-type symptoms). Due to the sustained, constant GH elevation, these observations can be more pronounced than with short-acting GHRH analogs.
- Glucose metabolism: sustained elevated GH levels can raise fasting blood glucose and reduce insulin sensitivity in studies.
- Loss of pulsatility: the constant rather than pulsatile stimulation is discussed in the research literature as a trade-off against the natural GH rhythm.
- No long-term data: robust long-term human studies for research use do not exist. The profile rests primarily on short-term studies and anecdotal experience.
