Description

Clomifene (Clomiphene citrate, Clomid) is a first-generation selective estrogen receptor modulator (SERM). The substance is a mixture of two stereoisomers - Enclomifene (trans, antiestrogen) and Zuclomifene (cis, weakly estrogenic) in approximate 62:38 ratio. This mixture is important for the action profile and makes the difference from purer Enclomifene.

Per study reports Clomifene blocks estrogen receptors in the hypothalamus. This removes the negative feedback of estradiol on GnRH pulse frequency, and the hypothalamus increases GnRH secretion. The result is a rise in LH and FSH from the pituitary and secondarily a rise in endogenous testosterone production in the testes.

In recreational research practice Clomifene is a main component of post-cycle therapy (PCT) after anabolic or SARM research protocols when the HPG axis has been suppressed. Typical PCT protocols use Clomifene for 4-6 weeks, often combined with Tamoxifen in the early weeks.

Known effects in studies: visual tissue sensitivity (visual phenomena like halos, floaters - usually transient), occasional mood swings, in rare cases ophthalmological effects with longer use. Pharmacokinetics: oral, half-life of about 5 days (Zuclomifene portion) and about 14 hours (Enclomifene portion).
