Description

Enclomifene (Androxal) is the pure trans stereoisomer of Clomifene. While commercial Clomifene citrate is a mixture of Enclomifene (62%) and Zuclomifene (38%), Enclomifene contains only the antiestrogen-acting component. Zuclomifene has weak estrogenic residual action and accumulates due to very long half-life (about 5 days) in the body - exactly this component is omitted in Enclomifene.

Per study reports Enclomifene exerts the same SERM mechanism as the parent product: blockade of estrogen receptors in the hypothalamus, removal of negative feedback, rise of LH/FSH, and secondarily rise of endogenous testosterone production. The difference: cleaner stimulus without the slow-accumulating estrogen-like action of the Zuclomifene portion.

Repros Therapeutics took Enclomifene through Phase III studies for treatment of secondary hypogonadism in the 2010s. Data showed Enclomifene normalized testosterone levels without the fertility-suppressing effect of exogenous testosterone - the sperm-supportive action was preserved.

Pharmacokinetically orally bioavailable with about 10-hour half-life - substantially shorter than Zuclomifene-containing standard Clomifene. This enables more precise dosing. Known side effects are similar to Clomifene, but visual phenomena (halos, floaters) are reported substantially less in studies than under the standard product.

In the recreational community Enclomifene is often positioned as “TRT alternative” or “bridge compound” between cycles.

Enclomifene sits in the mid range of the risk scale. As the pure trans isomer of Clomifene the compound is considered more cleanly tolerated in studies than the standard product, though the SERM profile remains.

Documented in studies and the literature:

  • Visual phenomena: halos and floaters are possible, but reported substantially less in the studies than under Zuclomifene-containing standard Clomifene.
  • Mood swings: described in the literature as a typical SERM accompanying effect.
  • Hot flashes: a possible accompanying effect of estrogen receptor modulation.
  • Shorter half-life: without the accumulating Zuclomifene portion, the action tapers off faster after discontinuation than with the standard product.
